tables or layers in design

I have been playing with different designs on sites and wanted to know a what some of you designers thoughts were. If I layout out a page in Layers instead of tables, what kind of problems could I run into. I was thinking that tables can be laid out in % instead of pixels so that that the page looks the same in 800x600 and also 1074x700. But with layers is their a way to do this...so far I have seen only pixel count in layers so if I design it in 800x600, it would look different in a larger screen?
Any thoughts here.

personally i would go for pixel instead of %
it looks exactly the same on all size
but just smaller and smaller in bigger and
bigger size ...
it it better to have to all stuffs spread out
on large size.
